Properties:
Sildenafil citrate is a modern therapeutic agent for oral treatment of erectile dysfunction. Physiologically penile erection is due to the release of nitric oxide at the cavernous body during sexual stimulation stage. Nitric oxide causes the activation of the enzyme guanylate cyclase that causes increased cGMP concentration that by relaxing the smooth muscle of corpus cavernosum allows increased blood flow to that level.
PROCOPS Sildenafil enhances the effect of nitric oxide by inhibiting the enzyme (phosphodiesterase type 5) responsible for degradation of cGMP in the corpus cavernosum, thus causing an increase in the concentration of cGMP produced by nitric oxide action on the stage sexual stimulation giving rise to smooth muscle relaxation and more blood, responsible for erection.
PROCOPS used at the recommended dose does not produce an erection in the absence of sexual stimulation.

Pharmacokinetics:
After oral administration is rapidly absorbed in the digestive tract with a bioavailability of 40% plasma peaking between 30 and 120 minutes.; The high-fat foods reduce these values. It has a wide distribution and plasma distribution. Its protein binding is 90%. It is metabolized in the liver by cytochrome P450 giving rise to its main metabolite N-desmethyl with an average life of approximately 4 hours. Dosage:

Contraindications:

Interactions:
Inhibitors of cytochrome P450 such as erythromycin, itraconazole, ketoconazole may increase levels of sildenafil PROCOPS. Inducers of this system (rifampin) decrease its concentration. Administered with nitroprusside hypotensive effects of the latter. Cimetidine increases the concentrations of sildenafil.
